It all starts with someone telling you a short, common story. Stand-up comedians call this a setup. Now, you're supposed to think about this story in a certain way. Your brain makes connections from this setup to a punch-line that you think will follow. You definitely won't be hearing the same punch-line you had in mind, but the other one, which you'll laugh at. Chances are you still don't quite understand this, so the following example should explain it better.

The Setup:
What is the most satisfying thing next to being shot at and missed?
The Punch-line:
An income tax refund.

What did you think when you heard the setup? I am sure you have thought that the car is being driven by one of the three people. It's normal to think that way, as that is just the way our brain was made to work. But as I, previously said, you didn't expect this kind of punch-line. Be sure to remember this, as this is the main principle of humor.

A good setup is of great importance. A good comedian needs a good setup which delivers only the basic facts (so that there are many possible ways of thinking), so that the audience would have to think hard and be convinced that their way of thinking is the right one. The more you think that your punch-line is the most suitable one, the more you'll laugh when you hear the actual one. But if you're already familiar with the punch-line the joke won't be funny anymore as the brain won't be making any new routes.

This should be helpful in case you want to think of some brand new funny jokes. When someone tells a story that everyone thinks about in similar way, but you find out a different way to understand it. Now, if you tell this to others, it should be really funny.
